A multi-approach analysis of the genetic diversity in populations of Astyanax aff. bimaculatus Linnaeus, 1758 (Teleostei: Characidae) from Northeastern Brazil Neotropical Ichthyology
Pamponet,Vanessa de Carvalho Cayres; Carneiro,Paulo Luiz Souza; Affonso,Paulo Roberto Antunes de Mello; Miranda,Viviam Souto; Silva Júnior,Juvenal Cordeiro; Oliveira,Claudine Gonçalves de; Gaiotto,Fernanda Amato.
Few reports are available about the ichthyofauna of typical semi-arid rivers, although the regional diversity has been constantly threatened by human activities, mainly related to impoundment and construction of dams. The goal of the present work was to evaluate using different methods, the population genetic structure of a characin fish, Astyanax aff. bimaculatus, widespread throughout hydrographic basins of Bahia, Northeastern Brazil. Morphological (meristic and morphometric data), cytogenetic (karyotype and Ag-NOR), and molecular (RAPD and SPAR) analyses were carried out in specimens collected upstream and downstream of Pedra Dam, in the main channel of Contas River (Contas River Basin), and in the Mineiro stream, which belongs to the adjacent Recôncavo...

A remarkable autosomal heteromorphism in Pseudoryzomys simplex 2n = 56; FN = 54-55 (Rodentia, Sigmodontinae) Genet. Mol. Biol.
Moreira,Camila Nascimento; Di-Nizo,Camilla Bruno; Silva,Maria José de Jesus; Yonenaga-Yassuda,Yatiyo; Ventura,Karen.
Pseudoryzomys simplex, the false rice rat, is a monotypic genus of the Oryzomyini tribe (Sigmodontinae) distributed in part of Bolivia, Paraguay, Argentina and Brazil. Its diploid number has been described as 56 acrocentric chromosomes decreasing in size and no karyotype figure has been depicted. Herein, we present karyotypic data on P. simplex, including chromosome banding and molecular fluorescent in situ hybridization using telomeric sequences and the whole X-chromosome of its sister clade Holochilus brasiliensis (HBR) as probes. A case of remarkable autosomal heteromorphism due to the presence of a whole heterochromatic arm leading to the variability of FN is reported, as well as the occurrence of regions of homology between the X and Y chromosomes...

Abnormal meiotic behavior in three species of Crotalaria PAB
Ferreira,Kátia; Torres,Giovana Augusta; Carvalho,Isabelle Vilela de; Davide,Lisete Chamma.
The objective of this work was to compare the meiotic behavior and pollen grain viability of three species of Crotalaria. Slides for meiotic analysis were prepared by the air-drying technique. Pollen grain viability was measured by three staining procedures (Alexander's solution, tetrazolium chloride and fluorescein diacetate) and in vitro germination in a sucrose solution. Eight bivalents were observed, confirming previous reports on populations from other regions of Brazil, as well as from other countries. All species showed abnormal meiotic behavior as follows: in Crotalaria micans, cytomixis and abnormal chromosome pairing in diakinesis; in C. spectabilis, abnormal chromosome pairing in diplotene; in C. zanzibarica, shrunk nuclei in leptotene and...

American marsupials chromosomes: why study them? Genet. Mol. Biol.
Svartman,Marta.
Marsupials, one of the three main groups of mammals, are only found in Australia and in the American continent. Studies performed in Australian marsupials have demonstrated the great potential provided by the group for the understanding of basic genetic mechanisms and chromosome evolution in mammals. Genetic studies in American marsupials are relatively scarce and cytogenetic data of most species are restricted to karyotype descriptions, usually without banding patterns. Nevertheless, the first marsupial genome sequenced was that of Monodelphis domestica, a South American species. The knowledge about mammalian genome evolution and function that resulted from studies on M. domestica is in sharp contrast with the lack of genetic data on most American...

Biosystematic and chemosystematic studies in five South American species of Conyza (Asteraceae) Boletín de la Sociedad
Urdampilleta,Juan D.; Amat,Aníbal G.; Bidau,Claudio J.; Kolb Koslobsky,Nicolás.
The composition of the essentials oils of five species of Conyza Less from Argentina was determined by Gas Chromatography-Mass Spectrometer. This composition is associated to morphological and cytogenetic characters. The monoterpenes constitute more than 60% of the essential oils in C. blakei, C glandulitecta, C. sumatrensis var. sumatrensis and C. sumatrensis var. floribunda, in which limonene is the predominant compound. In C. bonariensis and C. primulaefolia the monoterpene content constitute less than the 40%. C. bonariensis presents only 13% limonene, while in C. primulaefolia it is absent. The similarity analysis of monoterpenes showed a relationship between the morphological and cytogenetic analysis, and the monoterpene content character seems to be...

Chromosomal analyses in Megalonema platanum (Siluriformes: Pimelodidae), an endangered species from South American rivers Neotropical Ichthyology
Carvalho,Rafael Augusto de; Sanchez,Sebástian; Swarça,Ana Claudia; Fenocchio,Alberto Sergio; Martins-Santos,Isabel C.; Dias,Ana Lúcia.
This study presents chromosomal data of Megalonema platanum from rio Tibagi, Paraná, Brazil and from rio Paraná, Argentina. The diploid number was equal 54 with karyotype composition of 24m+16sm+2st+12a in both populations. The AgNOR sites were detected in the terminal position of a submetacentric pair of the two analyzed populations, coinciding with secondary constrictions on the short arm of pair 15. CMA3 and FISH with 18S rDNA probe displayed fluorescent signals that correspond to the AgNOR sites and secondary constriction. The presence of a small acrocentric supernumerary chromosome can be observed in M. platanum from rio Tibagi, with centromeric heterochromatin. Others heterochromatic blocks were evidenced in the terminal position of some chromosome...

Chromosomal analysis of Centromochlus heckelii (Siluriformes: Auchenipteridae), with a contribution to Centromochlus definition Neotropical Ichthyology
Kowalski,Samantha; Paiz,Leonardo Marcel; da Silva,Maelin; Machado,Amanda de Souza; Feldberg,Eliana; Traldi,Josiane Baccarin; Margarido,Vladimir Pavan; Lui,Roberto Laridondo.
ABSTRACT Historically, there are divergences in the species allocation between Centromochlus and Tatia. This study aimed to generate the first cytogenetic data about Centromochlus and, by analyzing a population of Centromochlus heckelii from the Amazon River basin, to contribute as evidence to a historical taxonomic dilemma. Diploid number of 46 chromosomes and a heteromorphic pair was found in the female karyotypes, thus characterizing a ZZ/ZW sex chromosome system. Pale blocks of heterochromatin were located in centromeric regions of some chromosomes; however, the exclusive female chromosome (W) is almost entirely heterochromatic. AgNORs were detected in terminal position on the short arms of one acrocentric pair in males and two chromosome pairs in...

Chromosomal basis for a bilateral gynandromorph in Pycnogaster inermis (Rambur, 1838) (Orthoptera, Tettigoniidae) Naturalis
Barranco, P.; Cabrero, J.; Camacho, J.P.M.; Pascual, F..
Morphological and cytogenetic analyses of a spontaneous gynandromorph of Pycnogaster inermis, found in a natural population in the Spanish Sierra Nevada, have shown that the left side was male and the right side female, with almost perfect bilateral symmetry. Ventral valves of the ovipositor were very short, appearing as two spurs; dorsal valves were longer but still shorter than normal. The specimen had an apparently normal testis on the left side and a well-developed ovary on the right side. Cytogenetic analysis revealed the presence of 2n = 28 + X0 chromosomes in the testis and 2n = 28 + XX in the ovary, the same as standard males and females, respectively, of this species. This suggests that the gynandromorphism occurred at a very early developmental...

Chromosomal characterization of Amazonian freshwater stingrays with evidence for new karyomorphs and XX/XY sex chromosomes Genet. Mol. Biol.
Valentim,Francisco Carlos de Souza; Porto,Jorge Ivan Rebelo; Feldberg,Eliana.
Abstract Cytogenetic studies in the subfamily Potamotrygoninae have provided valuable insights into the understanding of the evolution and diversification of its species. In the present study, the chromosomal features of seven nominal potamotrygonin species are provided: Plesiotrygon iwamae (2n=74, FN=120), Potamotrygon amazona (2n=66, FN=107), P. constellata (2n=66, FN=110), P. leopoldi (2n=64, FN=102), P. motoro (2n=66, FN=106) from four different localities, and P. orbignyi (2n=66, FN=106), P. scobina (2n=66, FN=104), from Central Amazon. Additionally, we found a new karyomorph in P. wallacei. We considered the localization of Nucleolus Organizer Regions (NORs), as well as the pattern of constitutive heterochromatin, as species-specific characters. We...

Chromosomal differentiation in Kerodon rupestris (rodentia: caviidae) from the brazilian semi-arid region Mastozool. neotrop.
Lessa,Gisele; Corrêa,Margaret M. O; Pessôa,Leila M; Zappes,Ighor A.
Kerodon rupestris (Wied, 1820) is the most specialized species among caviid rodents and is endemic to the extensive rocky outcrops in the semi-arid region in Brazil. Herein we describe different karyotypes for K. rupestris, based on samples collected in Itapajé, Ceará (CE), Iraquara in Bahia (BA), and Botumirim in Minas Gerais (MG). Our samples included northern, center and southern populations in the range of the species distribution. The results were compared with a karyotype from Pernambuco previously described for the species. The diploid number (2n = 52) was constant among the analyzed population samples, but the fundamental number (FN) varied from 92 in Itapajé (CE) and Iraquara (BA), to 94 in Botumirim (MG). Pericentric inversions affecting one...

Chromosomal variability in the wild ornamental species of Symphysodon (Perciformes: Cichlidae) from Amazon Neotropical Ichthyology
Mesquita,Débora R.; Porto,Jorge I. R.; Feldberg,Eliana.
Cytogenetic studies were conducted on three discus species which inhabit the Amazon in Brazil: Symphysodon haraldi from Manacapuru, S. aequifasciatus from Tefé and S. discus from Barcelos. All individuals showed 2n=60 chromosomes, most of them biarmed. No sexual chromosomal heteromorphism was verified. However, different karyotypic formulae, owing to the presence of subtelocentric chromosomes, were verified for S. aequifasciatus and S. discus. One of the karyotypic formulae from S. aequifasciatus (cytotype 2) differs from the others, due to one of the homologues in the first chromosome pair being significantly larger than the other. A large variability was observed toward the nucleolar organizer regions (NOR) of S. haraldi and S. aequifasciatus. Although...

Chromosome analysis in Saccodon wagneri (Characiformes) and insights into the karyotype evolution of Parodontidae Neotropical Ichthyology
Nirchio,Mauro; Masache,Maria Cecilia; Paim,Fabilene Gomes; Cioffi,Marcelo de Bello; Moreira Filho,Orlando; Barriga,Ramiro; Oliveira,Claudio; Rossi,Anna Rita.
ABSTRACT Parodontidae is a relatively small group of Neotropical characiform fishes consisting of three genera (Apareiodon, Parodon, and Saccodon) with 32 valid species. A vast cytogenetic literature is available on Apareiodon and Parodon, but to date, there is no cytogenetic data about Saccodon, a genus that contains only three species with a trans-Andean distribution. In the present study the karyotype of S. wagneri was described, based on both conventional (Giemsa staining, Ag-NOR, C-bands) and molecular (repetitive DNA mapping by fluorescent in situ hybridization) methods. A diploid chromosome number of 2n = 54 was observed in both sexes, and the presence of heteromorphic sex chromosomes of the ZZ/ZW type was detected. The W chromosome has a terminal...

Chromosome comparison between populations of the collared peccary, Tayassu tajacu, raised in captivity Biocell
Carvalho de Souza,Patrícia; Salim Khayat,André; Chamon Seligmann,Igor; Rodríguez Burbano,Rommel Mario.
The collared peccary (Tayassu tajacu) is widely distributed over the American continent, being found from the south of the USA to the north of Argentina. In Brazil, it is spread all over the country, being one of the potential species to be raised in captivity. Therefore, the cytogenetic techniques could be a potential tool for reproductive monitoring of animals raised in captivity, mainly when destined for commercial purposes. This study had the objective of determining the chromosome number of two populations raised in captivity and characterizing them by GTG banding. For this purpose, an analysis was made of mitotic metaphases obtained from lymphocyte cultures made from blood samples of 11 animals, six of which from the Northeast and five from the North...

Chromosome loss in bi-parental progenies of tetraploid Pacific oyster Crassostrea gigas ArchiMer
Mccombie, Helen; Lapegue, Sylvie; Cornette, Florence; Ledu, Christophe; Boudry, Pierre.
Alterations of chromosome number have been observed in the somatic tissue of Crassostrea gigas diploids and artificial polyploids. Tetraploid oysters with abnormal chromosome numbers in some or all of their tissue are considered undesirable as parents either for triploids (produced via a cross with diploids and of aquacultural interest) or for tetraploid breeding. Aneuploid tetraploid oysters may confer the tendency to lose chromosomes and to revert to lower ploidy levels to their offspring. More directly, their offspring could have a lower ploidy because of potential links between somatic and gametic chromosome loss. The present study evaluated the phenomenon in six bi-parental tetraploid families bred from parents of differing somatic ploidy quality. The...

Chromosome mapping of ribosomal genes and histone H4 in the genus Radacridium (Romaleidae) Genet. Mol. Biol.
Anjos,Allison; Loreto,Vilma; Souza,Maria José de.
In this study, two species of Romaleidae grasshoppers, Radacridium mariajoseae and R.nordestinum, were analyzed after CMA3/DA/DAPI sequential staining and fluorescence in situ hybridization (FISH) to determine the location of the 18S and 5S rDNA and histone H4 genes. Both species presented karyotypes composed of 2n = 23, X0 with exclusively acrocentric chromosomes. CMA3+ blocks were detected after CMA3/DA/DAPI staining in only one medium size autosome bivalent and in the X chromosome in R. mariajoseae. On the other hand, all chromosomes, except the L1 bivalent, of R. nordestinum presented CMA3+ blocks. FISH analysis showed that the 18S genes are restricted to the X chromosome in R. mariajoseae, whereas these genes were located in the L2,S9 and S10...

Chromosome polymorphism in Ancistrus cuiabae Knaack, 1999 (Siluriformes: Loricariidae: Ancistrini) Neotropical Ichthyology
Mariotto,Sandra; Centofante,Liano; Miyazawa,Carlos S.; Bertollo,Luiz Antonio Carlos; Moreira Filho,Orlando.
Cytogenetic and FISH analyses were performed in 30 Ancistrus cuiabae specimens from a bay near the town of Poconé, in the Pantanal of Mato Grosso, Brazil. The observed diploid number was 2n = 34 chromosomes for both sexes and three distinct katyotypic formulae were found, namely cytotype A (20m, 8sm, 6st, Fundamental Number/FN = 68; 6 males and 11 females), cytotype B (19m, 8sm, 6st, 1a, FN = 67; 8 males and 4 females) and cytotype C (18m, 8sm, 6st, 2a, FN = 66; a single male). NORs's analyses showed that these regions were located in distinct sites on the NOR-bearing chromosome pair, according to cytotypes. Thus, in cytotype A, NORs were located in the terminal region of the short arm of the second metacentric chromosome pair; in cytotype B, they were...
